Dinoprostone Dosage

The specific dosage of dinoprostone a woman receives depends on how she responds to the medicine and whether she is given the vaginal insert, gel, or suppository. The various forms have different uses; however, all are inserted into the vagina. A healthcare provider will administer this product in a healthcare setting where emergency services are available if needed.

An Introduction to Your Dosage of Dinoprostone

The dose of dinoprostone (Cervidil®, Prepidil®, Prostin E2®) your healthcare provider recommends will vary, depending on a number of factors, including:

  • The particular dinoprostone product you are using
  • How you respond to the medicine.

As is always the case, do not adjust your dosage unless your healthcare provider specifically tells you to do so.

Dinoprostone Dosing Guidelines

The standard dinoprostone vaginal insert (Cervidil) dosage for preparing the cervix for labor and delivery is one vaginal insert, placed high into the vagina one time only. You will need to remain lying down for at least two hours after the insert is in place. The insert will be removed by your healthcare provider for any of the following reasons:

  • It is no longer needed
  • It has been in place for 12 hours
  • Active labor begins
  • Potentially serious side effects occur.

The standard dinoprostone vaginal gel (Prepidil) dosage for preparing the cervix for labor and delivery is 0.5 mg (the contents of one syringe) inserted one time into the cervix. You will need to remain lying down for at least 15 to 30 minutes after the medication is given. Additional doses may be given after six hours if there has not been an adequate response to the first dose. However, it is recommended that no more than three syringes (1.5 mg) be administered in a 24-hour period.

The standard dosage of dinoprostone vaginal suppository (Prostin E2) to abort a pregnancy or empty the contents of the uterus is one suppository (20 mg) inserted into the vagina, every three to five hours until the desired response is obtained. However, these suppositories should not be given for more than 48 hours. You will need to remain lying down for at least 10 minutes after the suppository is placed.
